Lipocalin-2 (LCN2) is a secreted glycoprotein involved in innate immunity that has roles in iron homeostasis, infection, and inflammation. It was originally identified as a major constituent of the neutrophil secondary granules, so it is also known as neutrophil gelatinase-associated lipocalin (NGAL).
It is highly expressed in the central nervous system in response to injury and inflammation and has also been identified as a potential marker for Alzheimers disease. Increased levels of Lipocalin-2 have been seen in plasma from patients with mild cognitive impairment (MCI) and in CSF from patients with multiple sclerosis. Experimental models of Vascular dementia have shown that Lipocalin-2 mediates hippocampal damage and that decreased levels are associated with less white matter damage and cognitive decline.

The human Lipocalin-2 Kit is a sandwich ELISA. The capture antibody is a polyclonal human Lipocalin-2 antibody pre-coated onto the 96-well strip plates provided in the kit. Human test samples and standards of known Lipocalin-2 concentration are added to these wells and allowed to complex with the bound Lipocalin-2 antibody. A biotinylated human Lipocalin-2 polyclonal antibody is then added. This detection antibody binds to the antigen thus completing the sandwich. After washing, an enzyme Avidin-Biotin-Peroxidase complex (ABC) is added which binds to the second antibody. The peroxidase substrate TMB is added to induce a coloured reaction product. The intensity of this coloured product is directly proportional to the concentration of Lipocalin-2 present in the samples. The purpose of this kit is the in-vitro quantitative determination of human Lipocalin-2 in samples such as sera, plasma, tissue lysates and cell culture supernates. This kit has been configured for research use only and is not to be used in diagnostic or clinical procedures.

Iron-trafficking protein involved in multiple processes such as apoptosis, innate immunity and renal development. Binds iron through association with 2,5-dihydroxybenzoic acid (2,5-DHBA), a siderophore that shares structural similarities with bacterial enterobactin, and delivers or removes iron from the cell, depending on the context. Iron-bound form (holo-24p3) is internalized following binding to the SLC22A17 (24p3R) receptor, leading to release of iron and subsequent increase of intracellular iron concentration. In contrast, association of the iron-free form (apo-24p3) with the SLC22A17 (24p3R) receptor is followed by association with an intracellular siderophore, iron chelation and iron transfer to the extracellular medium, thereby reducing intracellular iron concentration. Involved in apoptosis due to interleukin-3 (IL3) deprivation: iron-loaded form increases intracellular iron concentration without promoting apoptosis, while iron-free form decreases intracellular iron levels, inducing expression of the proapoptotic protein BCL2L11/BIM, resulting in apoptosis. Involved in innate immunity, possibly by sequestrating iron, leading to limit bacterial growth.
