T-cell intracellular antigen 1 (TIA1) is an RNA binding protein that binds to the 5'TOP sequence of 5'TOP mRNAs. Under stress conditions, TIA1 localises to cellular RNA-protein conglomerations (stress granules). The major granule-associated species is a 15-kDa protein that is likely derived from the carboxyl terminus of the 40-kDa product by proteolytic processing.
TIA1 has been shown to co-localise with hyperphosphorylated tau and progressively accumulate with aggregated tau in tau-mediated neurodegeneration. Additionally mutations in the TIA1 gene have been associated with amyotrophic lateral sclerosis, frontotemporal dementia and Welander distal myopathy.
