The vacuolar protein sorting 35 (VPS35) is a major component of the retromer recognition core complex which regulates intracellular protein sorting and trafficking.
Mutations in the VPS35 gene are linked to late-onset, autosomal dominant familial Parkinsons disease (PD) , which may involve a link with tau pathology with VPS35 shown to be significantly reduced in two distinct primary tauopathies (Progressive Supra-nuclear Palsy and Picks disease, two distinct primary tauopathies) [1]. There may also be a link between VPS35 and ?-synuclein accumulation, but more work is required to establish the details of the pathiogenic/neuroprotective role of VPS35.
