Profilin 1 (PFN1) is a member of the profilin family of small actin-binding proteins. This 140-amino-acid protein acts as a major regulator of actin polymerisation through its binding of actin-ADP monomers, promoting the conversion of actin-ADP to actin-ATP. PFN1 also binds to phosphoinositides and a large network of proteins with poly-L-proline stretches. Through these binding interactions, PFN1 regulates a number of cellular functions including actin dynamics, membrane trafficking, neuronal synaptic structure and activity and small GTPase signaling.
Mutations in the PFN1 gene are associated with amyotrophic lateral sclerosis (ALS).

Binds to actin and affects the structure of the cytoskeleton. At high concentrations, profilin prevents the polymerization of actin, whereas it enhances it at low concentrations. By binding to PIP2, it inhibits the formation of IP3 and DG. Inhibits androgen receptor (AR) and HTT aggregation and binding of G-actin is essential for its inhibition of AR.
